Proteinase K, Molecular Biology Grade (P8107) can be used with T2010 as a direct substitute for Monarch Proteinase K (T2001). The Proteinase K Reaction Buffer (T2003) is not needed with P8107, so proceed without this buffer.
NEB offers another DNase I (RNase-free) (M0303) and DNase I Reaction Buffer (10X) (B0303) that can be used in place of Monarch DNase I (T2004) and Monarch DNase I Reaction Buffer (T2005). For an on-column DNase I treatment, mix 10 µl of DNase I (RNase-free) (M0303) and 70 µl of DNase I Reaction Buffer (10X) (B0303) per reaction and proceed as indicated in the protocol. For an in-tube DNase I treatment, directly substitute 5 µl of the M0303 DNase I (RNase-free) and DNase I Reaction Buffer (10X) and proceed.
