For some samples with very high DNA content, such as spleen tissue, adding ethanol to the flow-through leads to precipitation of genomic DNA. Loading this insoluble precipitate onto Column S2A can lead to increased DNA contamination in the final eluate. We recommend not loading this precipitate to Column S2A. Note that this insoluble precipitate is different from the transient cloudy appearance, which disappears with mixing and has no impact on performance.
