Cleavage of ssRNA with RNase If (NEB #M0243)
1. Set-up the reaction as follows:

| Single stranded RNA |
0.1 to 0.5 μgs |
| NEBuffer 3 (10X) |
2 μl (1X) |
| Nuclease-free H2O (NEB #B1500) |
up to 20 μl |
| RNase If |
1 μl (50 units) |
2. Mix thoroughly.
3. Incubate at 37°C for 15 minutes.
4. Heat Inactivation 70°C for 20 minutes.
