The accompanying library preparation protocols for sequencing NEBNext RSV amplicons use diluted RSV amplicons as inputs for library prep. However, if a cleanup of amplicon cDNA is preferred, 0.8X cleanup with SPRIselect or AMPure beads is recommended. For example, if amplicon cDNA volume is 25 µl, 20 µl (0.8X) of SPRIselect/AMPure beads would be used to bind the cDNA, followed by two washes with 200 µl of 80% EtOH, and elution in 15-30 µl of water.
