No, the aptamer-based hot start mechanism does not require an extended initial incubation. The aptamer/inhibitor is released from the polymerase during normal cycling conditions, so Standard Taq protocols can be used. We recommend incubating at 95°C for 30 seconds for the initial denaturation step.
