DNA for Ion Torrent™

In general, library preparation of fragmented DNA for use with the Ion Torrent™ sequencing platform involves repair of 3′ and 5′ ends before ligation to an adaptor. After ligation, adaptor-DNA constructs are purified and size selected, and amplified via polymerase chain reaction (PCR). For more information about DNA Library Construction, choose the workflow tabs below.

Libraries prepared in this manner are suitable for template preparation, the next step in the Ion Torrent sequencing workflow.

NEBNext® reagents are available for each step in the DNA library preparation workflow for the Ion Torrent™ platform. For DNA library preparation, reagents from NEB are available with or without reagents for enzyme-based fragmentation. Each of these reagents has been functionally validated by preparation of a genomic DNA library followed by Ion Torrent sequencing. For representative results of both library construction using NEBNext and Ion Torrent run results, choose the tabs below.

Representative Results from Libraries Constructed Using NEBNext for Ion Torrent

0.5 µg of DNA from 3 different genomes with varying GC content were used to construct 200 bp and 400 bp libraries using the NEBNext Fast DNA Fragmentation and Library Prep Set for Ion Torrent, analyzed by the Agilent® Bioanalyzer®.
Respresentative Ion Torrent Run Report
A typical Ion Torrent run report for libraries made from E. coli (K12 MG1655 strain) genomic DNA using NEBNext Fast DNA Fragmentation & Library Prep Set for Ion Torrent. A. Alignment Summary. 72 Mb of data was generated, with an average genome coverage of 15.5 X, from approximately 0.5 Million 200 bp reads. B. Read length histogram from 200 bp run.
