Description: This synthetic peptide, Autocamtide-2 Related Inhibitory Peptide (AIP), is a highly specific and potent inhibitor of Ca2+/Calmodulin-dependent Protein Kinase II (CaMKII). AIP is a non-phosphorylatable analog of Autocamtide-2, a highly selective peptide substrate for CaMKII (NEB #P6062). AIP inhibits CaMKII with IC50* of 100 nm. The AIP inhibition is non-competitive with respect to Syntide-2 (CaMKII Peptide Substrate, NEB #P6059) and ATP but competitive with respect to Autocamtide-2. The inhibition is unaffected by the presence or absence of Ca2+/calmodulin. 1 µM AIP completely inhibits CaMKII activity, but did not affect PKA, PKC and CaMKIV (1).
* IC50 is a concentration of the inhibitor required to obtain 50% inhibition.
Amino Acid Sequence: KKALRRQEAVDAL (Lys-Lys-Ala-Leu-Arg-Arg-Gln-Glu-Ala-Val-Asp-Ala-Leu)
Purity: AIP is > 95% pure as determined by HPLC and mass spectrometry.
Suggested Working Concentration:Dilution of the supplied solution (10 mg/ml = 6.7 mM) to a final concentration of 1 µM is recommended.
