  • Recombinant fusion protein (E. coli)
  • General protein kinase substrate
  • Suitable for mass spectrometry, Western blot, dot blot and ELISA assays
  • No basal phosphorylation
Description:
Myelin Basic Proteinf (MyBPf, isoform 4) is expressed as a 29 kDa fusion protein with a Chitin Binding Domain at its N-terminus, and a 6x His tag at its C-terminus. MyBPf can be used as a substrate for kinases, such as MAP Kinase, cAMP-dependent Protein Kinase, Calmodulin-dependent Protein Kinase, Abl Tyrosine Kinase, etc. (1-4). It has no basal phosphorylation or post-translational modifications. The phosphorylation of MyBPf can be analyzed using a variety of techniques, including western blot, array, dot blot, radioisotope analysis or mass spectrometry. It can also be used in ELISA assays by coating 96-well plates followed by a kinase reaction and phospho-specific antibody detection (Fig. 1).

This pack is suitable for at least one 96-well ELISA.

Reaction Conditions: Reaction can be carried out according to the kinase protocol suggested by the manufacturer.

Figure 1, ELISA Assay: Phosphorylation of recombinant MyBPf and native bovine MyBP by different protein kinases examined by a phospho-specific antibody. Recombinant MyBPf has low background whereas bovine MyBP has high noise to background ratio.

Source:
Isolated from a strain of E. coli that carries a clone expressing the MyBP fusion protein under a T7 expression system.
