 Description: The error rate of Phusion™ High-Fidelity DNA Polymerase in HF Buffer (4.4 x 10-7) is lower than that in GC Buffer (9.5 x 10-7). Therefore, the HF Buffer should be used as a default for high fidelity amplification. However, GC Buffer can improve the performance of Phusion DNA Polymerase on some difficult or long templates, i.e. GC rich templates or those with complex secondary structures. Use of the GC Buffer is recommended for those instances where amplification with HF Buffer has failed.
The detergent-free buffers for Phusion DNA Polymerase are designed for applications like DHPLC where PCR products are recommended to be free of detergents. They can also be used in microarray applications, as detergents can cause foaming when PCR products are spotted on microarray slides.
