Q1: Why does AfeI's catalog number start with V instead of R?

A1: This enzyme is made by SibEnzyme. As a service to our customers, we occasionaly supply enzymes with unique specifications that are exclusively produced by another supplier.


Q2: What buffers can be used for double digests?

A2: NEBuffer 4 is very similar in composition to the supplied SEBuffer Y and gives 100% activity. NEBuffer 2 can also be used if the amount of AfeI is doubled. We do not know the activity the activity of NEB enzymes in the SEBuffer system.


Q3: Does AfeI replave one previously sold?

A3: Yes, this enzyme replaces Eco47III (ER0321). AfeI recognizes the same sequence AGCˆGCT, and cleaves at the same position as Eco47III.


Q4: Are more units of AfeI required to cut supercoiled DNA than lambda DNA?

A4: Yes, 10 units are required to cleave 1 μg pBR322.
