Q1: Do BmrI generated ends ligate well?

A1: BmrI produces DNA fragments that have a single-base 3´extension which are more difficult to ligate than blunt-ended fragments. After 2-fold overdigestion with BmrI, >85% of the DNA fragments can be ligated. Of these, >95% can be recut.


Q2: Is extended digestion of BmrI recommended?

A2: Extended digestion is not beneficial because the enzyme is not stable in reaction.
