Q1: How do you recommend using BpmI?

A1: 1. Do not digest for more than one hour.
2. If attempting to ligate after cleavage, then phenol extract first to remove BpmI bound to DNA ends.
3. When ligating, treat as you would a blunt- ended ligation. Two base pair overhangs do not ligate easily.


Q2: Is BpmI blocked by overlapping dcm methylation?

A2: No.


Q3: Is extended digestion of BpmI recommended?

A3: Extended digestion is not beneficial because the enzyme is not stable in reaction.
