Q1: What factors inhibit AgeI?

A1: AgeI is blocked by CpG methylation.


Q2: Are extended digests of AgeI recommended?

A2: No, AgeI loses activity rapidly in a reaction. For extended digests use room temperature. AgeI is 100% active and more stable in reaction at 25°C. Adding 100 μg/ml BSA has also been shown to increase stability of some enzymes during extended reactions.


Q3: What is the reaction temperature of AgeI?

A3: The reaction temperature is 37°C. NEB previously suggested 25°C,however, cloning allowed us to provide a more robust preparation that can be used at 37°C. It is still 100% active at 25°C and this temperature is recommended for extended digests.
