BcgI FAQ

See the Restriction Endonucleases FAQ also.

Q1: Why isn't BcgI cutting my DNA?
Q2: Does BcgI require SAM for activity?
Q3: Do BcgI generated ends ligate?
Q4: What is the molecular weight of BcgI?

Q1: Why isn't BcgI cutting my DNA?

A1: BcgI requires SAM for activity. SAM is only stable for six months when stored at -20°C. Fresh SAM can be purchased from us; ask for product number B9003S.

Q2: Does BcgI require SAM for activity?

A2: Yes, 20 µM SAM  is required for optimal activity.

Q3: Do BcgI generated ends ligate?

A3: BcgI excises a 32 bp fragment with 2-base 3´ overhangs. These overhangs are not specific, so whether the ends can be ligated depends entirely on the specific DNA sequence being cut.

Q4: What is the molecular weight of BcgI?

A4: The MW of BcgI is 180 kD estimated from PAGE.
