Q1: Are there any special considerations when working with BbvI?

A1: Like many type IIS (shifted cut site) restriction enzymes, BbvI seems to remain bound to its substrate after cleavage. Extended digests are not recommended because 1 unit is required to cleave 1 μg of DNA in 16 hours. Phenol extraction is the suggested method of DNA purification due to the binding properties of the enzyme.
