Q7: If a final concentration of 200 μM each dNTP is called for, how much of the mix should be added to a 100 μl reaction?

Q1: What is supplied with the Deoxynucleotide Solution Mix?

A1: Each vial contains 8 μmol in a volume of 0.8 mls. A small pack contains one vial and a large pack contains 5 vials.

Q2: What is in the Deoxynucleotide Solution Mix?

A2: An equimolar mixture of ultrapure dATP, dCTP, dGTP, and dTTP.

Q3: How stable are the nucleotides?

A3: When stored at -20°C, they are stable for years. If they are freeze thawed frequently they should be stable for a year. If the solutions are stored at 4°C bacteria can grow in them. Bacterial growth is accelerated if the solution is diluted in a buffer containing magnesium. Discard solutions stored at 4°C after one week.

Q4: What is the concentration of Deoxynucleotide Solution Mix?

A4: The concentration is 10 mM of each dNTP.

Q5: What polymerases can the Deoxynucleotide Solution Mix be used with?

A5: Any DNA Polymerase from NEB.

Q6: How does this product differ from the Deoxynucleotide Solutions Set (NEB# N0446)?

A6: The Deoxynucleotide Solutions Set contains four vials, one containing dATP, dCTP, dGTP, and dTTP. The concentration of each is 100 mM. If equal volumes of nucleotides are mixed from the Deoxynucleotide Solutions Set the concentration would be 25 mM each dNTP.

Q7: If a final concentration of 200 μM each dNTP is called for, how much of the mix should be added to a 100 μl reaction?

A7: Add 2 μl. Determined by the equation 100 μl x 200 μM = x μl x 10 mM. Converting gives 100 μl x 200 μM = x μl x 10,000 μM. Solving the equation gives x = 2 μl.
