Exonuclease T FAQ

See the RNA Reagents FAQ also.

Q1: Will Exonuclease T degrade single stranded RNA?
Q2: Can Exonuclease T be heat inactivated?
Q3: What is the activity of Exonuclease T in the NEBuffers?
Q4: What is the molecular weight of Exonuclease T?
Q5: Does Exonuclease T work as well as DNA Polymerase I, Large (Klenow) Fragment (NEB# M0210) for 3' extension removal?
Q6: How is Exonuclease T different from Exonuclease I (NEB# M0293)?

Q1: Will Exonuclease T degrade single stranded RNA?

A1: Yes. Exonuclease T is also known as RNase T.

Q2: Can Exonuclease T be heat inactivated?

A2: Yes. Heat at 65°C for 20 minutes.

Q3: What is the activity of Exonuclease T in the NEBuffers?

A3: NEBuffer 1: 100%
NEBuffer 2: 75%
NEBuffer 3: 25%
NEBuffer 4: 100%

Q4: What is the molecular weight of Exonuclease T?

A4: 24 kD

Q5: Does Exonuclease T work as well as DNA Polymerase I, Large (Klenow) Fragment (NEB# M0210) for 3' extension removal?

A5: DNA Polymerase I, Large (Klenow) Fragment (NEB# M0210) or T4 DNA Polymerase (NEB# M0203) are preferred. They do, however, require the addition of nucleotides for 3' extension removal.

Q6: How is Exonuclease T different from Exonuclease I (NEB# M0293)?

A6: Exonuclease I will not remove short 3' extensions.
